Course Description:
Conduct a survey of the "Natural Area" for entomopathogenic nematodes (EPNs), and collect bark beetles for the lab. Set up traps. (Students will stop by the lab to check for emergence of nematodes every day) .
Observe different forms of nematodes from different insects: Beetles, cockroaches, crickets, Galleria mellonella (from baiting soil).
Galleria mellonella rearing
Nematode rearing. Collect Galleria from USDA. Each group of students set up a chamber to rear nematodes. Steinernema riobrave and S. glaseri will be used. (Student presentation).
Pathogenicity test: Galleria, fall armyworms, cut worms.
Nematodes used: S. riobrave, S. glaseri. (Student presentation).
Life cycle study: Dissect Galleria 1, 2, 3, 4 days after being exposed to S. glaseri. Look under a compound microscope. Described nematode stages observed. (Student presentation).
Survival test: Release nematodes in soil of 5 containers, and check survival after 2 weeks by baiting technique. (Student presentation).
Identification . Get results for survival test. (Student presentation).
DNA extraction. (Student presentation).
PCR process
Purify DNA for sequencing. RFLP with Alu I.
Working with DNA sequences

Grading: : Final grade will be based on student’s work in the lab and their notebook.
